Description
The dataset contains the raw files of the LC-MS and HDX-MS experiments reported in the paper Jensen et al. Structure and dynamics of a promiscuous Xanthan Lyase from Paenibacillus nanensis and the design of variants with increased stability and activity. The HDX-MS data in the paper serves the purpose of identifying the areas within the studied protein that display significant changes upon removal of the natively-present calcium atoms within the protein. By comparing the HDX of the protein in its native state and in the presence on 5 mM of the chelator EDTA, the regions impacted by Calcium removal were identified. The identification of the affected regions by HDX-MS guided subsequent site-directed mutagenesis studies to increase the stability of the enzyme in the presence of chelators. Additional HDX data showed how that the protein can be refolded by the addition of CaCl2 after incubation with EDTA showing the perturbations caused by calcium removal are reversible. This dataset contains the following sets of raw files from Masslynx as .tar archives: 1. HDX-MS experiment on XL: XL and XL + 5mM EDTA states at 15s, 30s, 1min, 10min and 1h as well as the maximally labelled control (all replicates). 2. HDX-MS experiment on XL: Single 10min timepoint for native XL, XL + 5mM EDTA and XL + 5mM EDTA + 10mM CaCl2 3. LC-MS data related to the sugars release by PXL when in presence of the Xanthan gum substrate.
